چکیده

The Alzheimer’s disease (AD) is the most frequent form of dementia worldwide. The major neuropathological hallmarks of the disease are loss of neurons and synapse, senile plaques (extracellular aggregates primarily composed of ß-amyloid; Aß) and neurofibrillary tangles (aggregates of hyperphosphorylated forms of the microtubule-associated tau protein) throughout cortical and limbic regions of the brain. The definite diagnosis still requires histopathological conformation according to the criteria, however, in recent years substantial progress has been made in the area of early biomarker development. The use of cerebrospinal fluid as a testing platform is very promising because the CSF protein composition can reflect the pathological processes of the brain and because it is easily accessible by a lumbar puncture. Some proteins and peptides such as ß-amyloid 1-42 (A┚142), ß-amyloid 1-40 (A┚1-40), total tau (tau) and hyperphosphorylated tau (p-tau) have been reported to meet the criteria for a biomarker. Another series of publications reported transthyretin, isoprostane, BACE1 activity and other proteins and enzymes as a potential biomarkers in AD. Whereas the biomarkers mentioned first have been studied extensively and were suggested to be included into clinical AD criteria, less information is available on the others. This review will focus on the importance of CSF based biomarkers in AD, covers the data available from the literature and highlights their role in the differential diagnosis of dementia.

The Participation of Three Brain Tissues in Alzheimer’s disease Diagnosis from Structural MRI

Alzheimer’s disease (AD) is a progressive and irreversible disease which gradually makes patients unable to do their daily routines. Although the present treatments can not cure the disease completely, its early detection can reduce symptoms and enhance the patients’ life quality. In the current literature, using the grey matter (GM) tissue which is known as an appropriate biomarker is highly c...

Reduced cerebrospinal fluid amyloid-b42 and increased retention of florbetapir positron emission tomography are biomarkers reflecting cortical amyloid load in Alzheimer’s disease. However, these measurements do not always agree and may represent partly different aspects of the underlying Alzheimer’s disease pathology. The goal of this study was therefore to test if cerebrospinal fluid and posit...

Nanotechnology for Alzheimer’s Disease Detection

Alzheimer’s disease (AD) is the most common cause of dementia among people of age 65 and older. The diagnosis of sporadic AD is based on clinical exclusion criteria and is only definite at necropsy. So, biochemical markers for AD would be of great value for its early diagnosis. During the last decade, research efforts have focused on developing cerebrospinal fluid (CSF) biomarkers for AD. The d...

New CSF biomarkers on the block

A growing number of cerebrospinal fluid biomarkers are now available to capture different aspects of Alzheimer’s disease. People are increasingly aware that these biomarkers represent a real-time reflection of pathological mechanisms that are ongoing in the brain. These novel markers can be added to the panel of existing ones like amyloid beta, total Tau and phosphoTau that are currently used f...

Modeling of Pathological Traits in Alzheimer’s Disease Based on Systemic Extracellular Signaling Proteome*□S

The study of chronic brain diseases including Alzheimer’s disease in patients is typically limited to brain imaging or psychometric testing. Given the epidemic rise and insufficient knowledge about pathological pathways in sporadic Alzheimer’s disease, new tools are required to identify the molecular changes underlying this disease. We hypothesize that levels of specific secreted cellular signa...
